Using various preclinical models and patient data, the authors demonstrate that these inhibitors effectively block the active state of RAS proteins to trigger tumor regression and improved survival. The findings identify oncogenic flux—specifically through MYC expression and KRAS amplification—as a primary driver of drug resistance. Furthermore, the study reveals that combining these inhibitors with standard chemotherapy significantly enhances anticancer activity compared to single-agent treatments.
