This study investigates the shifting immune landscape in Alzheimer’s disease and cerebral amyloid angiopathy using advanced single-cell transcriptomics and spatial mapping. Researchers discovered that while microglia drive early neuroinflammation, CD8+ T-cells become the dominant immune responders as amyloid-beta plaques accumulate in the brain. A specific subset of these T-cells expresses interferon-stimulated genes, triggering a Type-I interferon response that recruits further immune cells via the CXCL10-CXCR3 signaling axis. These infiltrating T-cells cluster directly around plaques, where they undergo activation-induced exhaustion, a phenomenon also observed in human brain tissue.
