1163-Ecotypes and Archetypes of TNBC chemotherapy Response

This study presents a comprehensive single-cell and spatial transcriptomic atlas of early-stage triple-negative breast cancer (TNBC) to better predict patient responses to chemotherapy. By analyzing over 400,000 cells from treatment-naive patients, researchers identified four distinct cancer archetypes and thirteen internal metaprograms that characterize the diversity of tumor cells. The research further defines eight spatial "ecotypes," which are unique cellular communities formed by the interaction between malignant cells and the tumor microenvironment. Crucially, the data reveal that specific interferon signaling pathways and macrophage subtypes are strong indicators of a positive clinical outcome following treatment.
