1167-Regional Principles of Neuronal Protein Clearance
from Paper Talk
Researchers have developed a novel genetic tracing system to map how proteins produced in the brain are cleared through surrounding tissues. Unlike traditional injected tracers that often bypass the brain, this non-invasive ZsGreen reporter reveals that waste follows a "nearest exit" principle, where the anatomical origin of a protein determines its specific drainage route. The study identifies distinct immune niches in the skull and dura, such as tolerogenic B cells, that monitor these brain-derived antigens to maintain immune balance. Furthermore, the findings demonstrate that neuroinflammation and amyloid pathologies impair these pathways through different mechanisms, either by leaking proteins into the blood or trapping them within brain tissue. This comprehensive architecture suggests that spatial compartmentalization is fundamental to maintaining brain health and understanding regional disease vulnerability.References:Chayama Y, Rao NR, Perla D, Zhang Z, Reid M, Nelson S, Wen X, Ding B, Blumenfeld J, Apolonio A, Doddipalli S, Zhou H, Turhan SG, Shih PY, Brendel M, Fu YH, Ertürk A, Kolabas ZI, Huang Y, Yang AC.
