Researchers have developed D&D-seq, a novel single-cell technology designed to map DNA-protein interactions, even those that are weak or transient. By fusing a cytosine base editor to a nanobody, the method induces specific C-to-U mutations at sites where proteins like transcription factors are bound to the genome. This technique addresses the limitations of older methods that struggled with low-input samples or high-salt conditions that disrupted fragile molecular bonds. D&D-seq is highly versatile, integrating seamlessly with multi-omic workflows like scATAC-seq to co-profile chromatin accessibility and protein binding in individual cells. In practical applications, the authors used the tool to identify disrupted CTCF binding in primary mutant T cells and to predict 3D chromatin structures.
