1191-Spatially Single-Cell Atlas of Human Meningioma
from Paper Talk
This study utilizes spatially resolved single-cell analyses to map the complex cellular architecture of human meningioma, the most common adult brain tumor. By examining over 580,000 nuclei and millions of cells through spatial transcriptomics, the researchers identified specific transcriptional metaprograms that define both neoplastic and non-neoplastic cell states. A primary focus of the research is the tumor microenvironment, specifically how myeloid cell states evolve from the surrounding dura into the tumor and how these interactions influence tumor progression. The data reveal that while neoplastic cells exhibit significant regional diversity, chromosomal alterations (such as losses of 1p and 22q) remain relatively stable across different tumor areas. Ultimately, the authors demonstrate that these unique non-neoplastic cell states can refine clinical outcome predictions beyond traditional molecular grouping and WHO grading. These findings provide a high-resolution atlas intended to inform future therapeutic strategies for managing meningioma heterogeneity.References:Landry A P, Yefet L S, Wang J Z, et al.
