1214-Prognostic Atlas for Pancreatic Cancer
from Paper Talk
Tang et al. establish a high-resolution prognostic atlas for pancreatic cancer by integrating single-nucleus transcriptomes with survival data from 152 patients. This study shifts the paradigm from traditional bulk-level analysis to cell-type-specific resolution, allowing researchers to link individual cell behaviors to clinical outcomes. The authors developed ctPANDA, an interactive web platform that empowers the scientific community to explore these transcriptomic profiles and discover new biomarkers. Using single-cell spatial transcriptomics, they mapped over three million cells to identify how cellular neighborhoods and spatial proximity influence therapeutic responses. A key breakthrough of the research is the identification of PLOD2 as a pan-cell-type therapeutic target consistently associated with poor prognosis. To demonstrate clinical potential, they synthesized a PROTAC-G3 compound that successfully degrades PLOD2 and inhibits tumor progression in preclinical models.References:Tang R, Jin L, Chen C, et al.
