This research identifies MARCKS as a critical driver of hepatocellular carcinoma (HCC) progression by reprogramming the tumor immune microenvironment. Utilizing multi-omics analysis, the study demonstrates that elevated MARCKS expression correlates with aggressive clinical features and poor patient outcomes. The protein is primarily enriched in myeloid cells, where it facilitates the transition of macrophages into an immunosuppressive M2-like state. Mechanistically, this process is mediated through the activation of the JAK/STAT3 signaling pathway, which fosters immune evasion. Experimental validation shows that silencing MARCKS reduces tumor growth and limits the development of these pro-tumorigenic immune cells.
