While the lipid phosphatidylserine (PS) typically stays inside cell membranes, neurotoxic stress causes it to cluster at dendritic branching points, a process driven by the loss of Atp8a2 expression across various neurodegenerative models. Crucially, the study distinguishes between the protein’s enzymatic activity and its physical presence; while inhibiting its activity causes PS exposure, only the reduction of total Atp8a2 levels triggers architectural collapse and cell death. The findings suggest that Atp8a2 maintains neuronal integrity through interactions with the cytoskeleton, independent of its lipid-shuffling function.
