This research article investigates the role of the miR-424(322)~503 cluster in the development of colorectal cancer triggered by PTEN deficiency. Using genetically modified mouse models, researchers discovered that while this microRNA cluster acts as an oncogene in other tissues, it functions as a tumor suppressor in the colon. The study found that mice lacking both PTEN and the microRNA cluster developed more numerous and aggressive tumors, characterized by increased dysplasia and adenocarcinoma. Mechanistically, the loss of these microRNAs leads to the hyperactivation of the MAPK and TGFβ signaling pathways, as well as the derepression of the growth factor IGF1. Evidence from human patient data further supports these findings, showing that the cluster is often downregulated in human colorectal tumors.
