1 source·Aug 10, 2026This research identifies interleukin 17 receptor A (IL17RA) as a novel, direct ligand for the Vδ7 γδTCR in mice, a discovery that clarifies long-standing mysteries regarding γδT cell antigen recognition. Using cDNA library screening and surface plasmon resonance, the authors demonstrate that IL17RA interacts with germline-encoded regions of the Vδ7 chain rather than traditional CDR3 loops. This nonclonotypic interaction is essential for the development and survival of a major population of TH1-like effector γδT cells found in secondary lymphoid organs. The study proves that IL17RA is necessary for thymic selection, acquisition of effector functions, and the homeostatic proliferation of these cells in the periphery.
