This research article identifies the CARM1 epigenetic enzyme as a primary negative regulator of cDC1 dendritic cells, which are essential for initiating anti-tumor immunity. The authors demonstrate that inactivating the Carm1 gene or using a small-molecule inhibitor significantly improves the ability of these cells to cross-present cancer antigens to cytotoxic T cells. Mechanistically, CARM1 expression is triggered by the immunosuppressive cytokine TGFβ, which limits the chromatin accessibility of genetic pathways required for dendritic cell activation. By removing this barrier, the researchers observed increased T cell infiltration and enhanced effectiveness of PD-1 blockade in treating resistant tumors.
