Using advanced multimodal spatial profiling and single-nucleus sequencing, researchers identified that different cognitive impairments arise from unique biological processes rather than a single disease mechanism. Specifically, language and fluency deficits were linked to widespread inflammatory responses in the glia and blood vessels across the brain. In contrast, executive dysfunction was associated with localized synaptic and mitochondrial failures specifically within the deep layers of the neurons. These findings suggest that the clinical variety of ALS is driven by phenotype-specific interactions that are not fully explained by traditional protein pathology.
