This research article explores how activating the 5-HT2AR serotonin receptor can significantly enhance antitumor immunity in colorectal cancer. The study identifies that while LSD suppresses tumor growth, its use is limited by psychedelic effects caused by brain penetration. To overcome this, researchers developed IHCH-8110, a specialized non-brain-penetrant agonist that selectively targets receptors in the peripheral nervous system. This compound works by activating enteric glial cells, which then release specific cytokines to recruit and empower CD8+ T cells within the tumor. Notably, this treatment can transform "immune-cold" tumors into responsive ones, markedly increasing the effectiveness of anti-PD-1 immunotherapy.
