Demi menggapai dunia baru penghasil rempah-rempah, Portugis melakukan Ekspedisinya hingga ke Timur Nusantara. Setelah menaklukan Goa di India, Alfonso de Albuquerque menaklukan Malaka di tahun 1511 dilanjutkan dengan menjalin persahabatan dengan raja-raja di Timur Nusantara. Kekuasaan Portugis di Malaka dan Kepulauan Maluku tidak dapat bertahan lama dan hanya menyisakan koloni di Timor Portugis.
