This research article introduces a novel antibody-drug conjugate (ADC) designed to treat T cell cancers by targeting the TRBC2 constant region of the T cell receptor. Unlike existing treatments that deplete all T cells and cause severe immunosuppression, this therapy exploits the fact that individual T cell malignancies express only one of two possible alleles, either TRBC1 or TRBC2. By engineering a high-affinity antibody named JX1.1, the researchers successfully targeted cancerous cells while preserving a significant portion of healthy T cells to maintain immune function. Laboratory tests and animal models demonstrated that this ADC is effectively internalized by tumor cells and delivers a potent cytotoxic payload, leading to tumor regression.
