This research explores how human CD4+ T cell immunity is shaped by different stages and outcomes of Hepatitis C virus (HCV) infection. Using single-cell RNA sequencing and phenotypic profiling, the authors identified a stem-like subset of cells that emerges early and sustains the immune response during chronic infection. While successful antiviral therapy (DAA) leads to a long-lived memory pool, these cells retain a "chronic imprint" or molecular scar that distinguishes them from cells following spontaneous clearance. The study demonstrates that these stem-like cells are capable of recall activation, providing a framework for understanding T cell maintenance.
