NEJM July 2026 PARP and Androgen-Signaling Inhibition plus ADT in Metastatic Prostate Cancer

This study evaluates a novel treatment combination for patients with a specific form of advanced prostate cancer that remains sensitive to traditional hormonal therapy but carries genetic repair deficiencies. Researchers compared a regimen of the PARP inhibitor talazoparib paired with enzalutamide against a standard control group to measure how long patients lived without their cancer worsening. The results demonstrated that adding talazoparib significantly delayed disease progression and improved survival rates at the three-year mark, particularly for those with certain genetic mutations. While the combination proved highly effective at slowing the spread of the disease, it also led to a higher frequency of serious side effects, most notably severe anemia.
