Nei neuroni, le cellule nervose che compongono il nostro cervello, svolge importanti azioni di regolazione della trasmissione sinaptica e di regolazione della barriera emato-encefalica. La beta amiloide è il risultato del clivaggio della proteina precursore dell’amiloide tramite l’azione di enzimi chiamati secretasi. Una volta prodotta in eccesso può accumularsi in forma solubile, aggregarsi in oligomeri, fino a precipitare formando placche amiloidi, che rappresentano una delle entità istopatologiche della malattia di Alzheimer. Recentemente, alcuni studi preclinici hanno evidenziato che la beta amiloide può indurre una attività neuronale anomala, e che crisi epilettiche possono incrementare la produzione di beta amiloide a loro volta, inducendo un circolo vizioso. A questi dati fa eco, dal punto di vista clinico, un’alta prevalenza di epilessia tra i pazienti con disturbi cognitivi, come anche di demenza nei pazienti epilettici. Facciamo un po’ d’ordine.
