Recientemente, un numeroso grupo de investigadores de varias universidades estadounidenses han generado una modificación del sistema CRISPR que permite afectar al funcionamiento de genes esenciales de las bacterias sin matarlas, y poder estudiar así mejor su función. Para ello, han conseguido una variante de enzima Cas que junto con un ARN complementario a un gen bacteriano puede unirse al mismo, pero sin cortarlo, es decir, sin destruirlo. La unión del ARN y del enzima Cas no cortante impide, sin embargo, el funcionamiento del gen en mayor o menor medida, por lo que la bacteria no puede tenerlo funcionando al cien por cien. Con el funcionamiento de uno o varios genes disminuido, podemos ahora analizar si estos afectan a la sensibilidad de una bacteria a un antibiótico frente al cual es resistente.
