Both are GLP-1 and glucagon dual agonists. Think of them like retatrutide minus the GIP component. And that glucagon piece is what makes them special.I go through the trial data for both compounds. We're talking up to 21% weight loss with mazdutide and 80% liver fat reduction. Survodutide earned FDA breakthrough therapy designation for MASH based on a 62% resolution rate. These are serious numbers.Here's what most people don't realize. These two peptides are NOT the same. Mazdutide runs close to a 50/50 split between GLP-1 and glucagon agonism. Survodutide is roughly 8 to 1 in favor of GLP-1. That changes everything about tolerability, appetite suppression, and how much metabolic burn you're actually getting.I also cover practical use cases. Cycling off reta without losing momentum. The “little blue pill” has a reputation as an erectile dysfunction drug, but when you actually look at the mechanism and the data, it’s doing way more than that.I’ve been a proponent of low-dose Cialis for a long time, and I still am. But lately I’ve been experimenting with rotating between Cialis and Viagra because I notice I can build a tolerance to Cialis over time. It still works, but I don’t feel the performance benefits as strongly unless I push the dose higher. What I’ve found is that going four weeks on low-dose Cialis, then four weeks on low-dose Viagra, seems to keep me more responsive overall while still giving me the benefits that matter for longevity.In this episode, I break down the real mechanism. Sildenafil is a PDE5 inhibitor that amplifies nitric oxide signaling and raises cGMP throughout the body. That means systemic vasodilation, improved endothelial function, better blood flow to organs, and anti-thrombotic effects through reduced platelet activation. And the downstream effects get even more interesting. Sometimes you’ll see it spelled Thymagen, but I’ve always preferred “Thymogen” because it just rolls off the tongue. Either way, we’re talking about a bioregulator peptide that works specifically on the immune system, and it’s one that’s been floating around research circles for a while… but I haven’t gone deep on it until now.Most of you have heard me talk about Thymalin — and I love Thymalin. I’ve used it a lot, and in certain use cases I even like it more than Thymosin Alpha-1. But here’s the key: Thymalin is broad-spectrum. It’s like a full thymus “reset.” Thymogen is more targeted — especially for the “post-viral” use case, that lingering sickness, the recovery phase, or even as a proactive tool when colds and flus are making their rounds. And honestly, that’s exactly why this episode matters right now, because I’ve been dealing with a little head cold myself after travel and lack of sleep.In this episode, I break down what Thymogen is (a synthetic dipeptide: L-glutamyl-L-tryptophan), why it’s interesting from a longevity perspective, and what we know about its safety history in Russia. Then we get into the mechanisms: T-cell maturation, immune surveillance, neutrophil function, and even some of the signaling pathways it appears to influence like ERK, mTOR-related proteins, STAT1, and chromatin remodeling. I also explain why I see Thymogen as more of an immune modulator than a blunt “stimulator,” which matters a lot if you’re dealing with inflammation or an overactive immune system.We’ll talk practical benefits — fewer infections, better recovery, inflammation control, and why the animal data is intriguing when you look at lifespan and tumor incidence. Then I’ll give you my straightforward protocol: 1–2 mg per day for 30–60 days, usually once or twice per year as a tune-up… and also how I’d deploy it if you’re actively sick or dealing with something that just won’t clear.Finally, I’ll compare Thymogen vs Thymalin, and I’ll explain how you can use them together: Thymalin as the foundation, Thymogen as the fine-tuning. We talk about the wild fact that DSIP can cross the blood–brain barrier, and why that makes nasal spray formulations surprisingly effective in real-world use.From there, we get into mechanisms: DSIP’s ability to lower ACTH and cortisol, act as a “stress-limiting” factor, nudge growth hormone a bit, modulate pain pathways, and influence neurotransmitters like Substance P.
